Ordering information
The number of index plates to order is dependent on your sequencing platform and protocol (reads per flow cell), the size of your panel and the desired read depth.
To multiplex samples in a single sequencing run, each sample must have a distinct combination of index sequences. Each 384-well Indexing Plate contains 384 distinct index combinations. The number of Indexing Plates to order for each flowcell is the sample multiplex per flowcell/384.
During the development of your Amp-Seq panel, your Biosearch Technologies team will provide guidance on the optimal Indexing Plates to use for your specific setup. Detailed guidance on sample multiplexing is available as an appendix in the Amp-Seq Reagent System manual available through your Biosearch Technologies representative.
Note: The index sequence file, required for demultiplexing, is provided by your Biosearch Technologies representative.
Examples
For a panel of 1,500 targets, sequencing at a desired depth of 150 reads per target on a NextSeq 550 instrument using a High Output Flowcell, the number of samples that may be multiplexed is (400,000,000/(1,500*150)) = 1,778 samples
To multiplex samples in a single sequencing run, each sample must have a distinct combination of index sequences. Each 384-well Indexing Plate contains 384 distinct index combinations. To multiplex the 1,778 samples in the example above, a total of 5 x 384-well index plates having different ‘Plate IDs’ will be required (1,778/384 = 4.63).
To run the 1,778 samples from the above example once, your Indexing Plate order should include 1 item of each of the below product codes:
