The I-TBHQ1 modification adds a non-fluorescent Black Hole Quencher®-1 dye internally or to the 5’ end of an oligonucleotide. BHQ®-1 labeled oligonucleotides are used in a wide array of applications, including dual-labeled fluorogenic probes for real-time PCR. The dye is conjugated via a fifteen atom linker to a modified thymidine which can participate in hybridization. The BHQ-1 dark quencher has strong absorption from 480 nm to 580 nm, which provides excellent quenching of fluorophores that fluoresce in this range, such as FAM, TET, CAL Fluor® Gold 540, JOE, HEX, CAL Fluor Orange 560, and Quasar® 570 dyes. BHQ-1 dye may quench fluorescence by both FRET and static quenching mechanisms.
