5'-DMT-dA(Bz) "Twist" Column; 1000 Å is a 10 µmol column packed with 5'-DMT-dA(Bz)-Suc-CPG, which is used for the placement of a deoxyadenosine (dA) base on the 3' end of the oligonucleotide sequence. The 5'-DMT-dA(Bz)-Suc-CPG support is linked to the CPG support through a succinyl linkage which can be cleaved in concentrated NH4OH or Methyl amine/ NH4OH(AMA). The dA has a benzoyl protecting group which is removed during the deprotection step using concentrated NH4OH or AMA. This support is compatible with fast deprotecting amidites and chemistry.

Product usage:

  • Synthesis conditions:

    Follow the instrument manufacturer's protocol, with noted changes, when using this product. When setting up the instrument for synthesis choose the 10µmol or 15µmol program for a successful synthesis using these columns

  • Cleavage conditions:

    Use concentrated ammonia for 30 minutes at 65 °C or 1:1 ammonia:methylamine (AMA) for 25 minutes at 25 °C.

  • Deprotection conditions:

    When using fast deprotecting amidites (eg. C-Ac, G-DMF) use concentrated ammonia for 1 hour or AMA for 30 minutes at 60 °C. When using standard amidites (eg. C-Bz, G-iBu) use concentrated ammonia for 5 hours at 60 °C.
