3'-DMT-dG(iBu)-Suc-CPG; 1000 Å is an inverse linkage CPG used to create a 3'-3' linkage on the end of an oligonucleotide. An inverted 3'-3' linkage is desirable when needing to block polymerase extension through the 3'-terminus of a PCR probe; additionally, oligonucleotides with inverted 3'-linkages are used for some sequencing techniques. The 1000 Å pore size is best suited for the synthesis of DNA sequences up to 100-mers in length.

Product usage:

  • Protecting group deblock conditions:

    3'DMT groups detritylate slowly. Increase detritylation time by 50%.

  • Cleavage conditions:

    Use concentrated ammonia for 90 minutes at 25 °C when using standard amidites or 1:1 ammonia:methylamine (AMA) for 25 minutes at 25 °C.

  • Deprotection conditions:

    Use concentrated ammonia for 5 hours at 60 °C.
